After clicking on the GP11-KB2731285-ENU.MSP I receive
Failed to Open XML file C:\Program Files\Microsoft Dynamics\GP2010\Dynamics.exe.config, system error -2147024786
There is no such directory so I do not understand where it is finding this path. Upgrade worked fine in an identical backup environment. GP2010R2 is installed in pathC:\Program Fiels\Microsoft Dynamics\GP2010R2

Typically this issues is going to be due to a registry entry eg. the registry entry is possibly pointing to the incorrect location for the GP Code folder.
During the service pack installation it is trying to open the Dynamics.exe.config file but it is failing to open because as an example, it may be looking for C:\Program Files (x86) folder and GP is installed in the C:\Program Files\Microsoft dynamics\GP2010 folder.
What you could consider doing is the following:
Rename the GP Code Folder from GP2010 to GP2010.OLD
Run a repair on Microsoft Dynamics GP 2010 in Program and Features
Once you run the repair it should create the Dynamics GP Code folder in the correct location eg: C:\Program Files (x86) which what you may find listed according to the Registry subsequently allowing you to try and install the service pack again.
