When a quote is assigned to another user, is it possible to have a custom field, like comments/reason, to capture details. In my limited understanding of the tool, we couldn't find any way of doing this through administration.
Thre is no standard configuration for this but you can customize/ confgiure your own implementation.
Create a new field (or use your existing field). Create a real time workflow and trigger it on Assign. Within the workflow, check if the reason field is populated, if not Stop the workflow and select Cancell. Under properties, display the error message.

As mentioned by Ravi Kashyap, there is no standard method to perform this. You can use custom code or workflow to perform this operation. You can create custom field to capture comments and use the custom code or workflow to prevent assignment if this new field is empty.
