When placing an order, my coworkers input it onto an excel spreadsheet, which I then retype onto the PO. Is there a way to upload the spreadsheet directly into the PO?

Integration Manager is likely to be your best option; an integration could be configured to read your spreadsheet (saved to CSV) and import the PO.
A macro is tricky when you have a scrolling window.
I agree with Harry, best tool for Integrations, we use it with all our customers and opens up lots more opportunities for importing.

There are numerous options. SmartConnect, eConnect, Integration Manager and Macros come to mind.
Smart Connect - Doesn't Require a GP User ($3000) to import Excel files right to GP.
eConnect - is powerful, but complicated.
Integration Manager is simple, and direct from Microsoft, but requires a GP User be logged in to run, so you have to factor this into the true cost of the solution.
Macros are free with GP, require a user be logged in, and need to be set up each time you are going to load data, but they have the added advantage of supporting custom fields when needed.
