One of our user can't make PO, even i have given all roles to create whatever, he wants?? Please check the above images for more clarification.

Did the assignment of the additional roles fix the issue or are these users still not able to create a PO?

Hi Shabir Ahmad ,
You can also try with below options in addition to whatever you have done so far at user role and privilege level.
1) Add Buying agent role to the user who is responsible to create purchase order in system.
2) Add Maintain purchase order details and Process purchase orders privileges to the user roles.

If you provided also the system administrator role, then I do agree with Mahim that is could be related to a personalization. The user can reset the personalizations on this form or you can reset his usage data.
Have you tried visible tab box tick under personalize ??
