I was able to give a user access to the extender and they have been using it, and then this morning they no longer are able to access it. It doesn't seem like any of their security and roles have changed. Any ideas?
They recently got updated to Windows 7...would that have made a difference at all?

It shouldn't make a difference but who knows - it could have.
Have you checked security? If you look up the following on a Security task it may help:
