Good morning 

is it possible for a PO of say between €500 and €1,000 goes for approval to manager A and PO exceeding €1000 and €5,000 goes for approval to manager B.

I know I can use the auto approval below the €500

    You can achieve this using the signing limit functionality.

    You can as well use conditions in your workflow setup.
