what difference for 'Exclude from Role' and 'Remove' in the assign user to role

In the [Assign users to Role], 
 
If i remove user role by "Exclude from role", the user seems to remove to specified roles. But its assignment mode is still "Manual"
 
May i know
1) what difference from 'Exclude from role' and 'Remove'
2) what is the purpose of assignment mode

    Exclude from Role temporarily removes a user from a role but keeps the assignment in the background, with the assignment mode still set to Manual Remove completely removes the user from the role and the assignment, and if re-assigned, the role assignment process starts fresh.
