If you are comfortable with writing codeunits you can try below solution :
You can send a test mail using CreateMessage and Send Functions in Codeunit 400(SMTP Mail) to check if your SMTP setup has been configured correctly.

As per Mohana its a nice functionality, but this functionality is not available in NAV 2013.
So follow the bellow link.
Test email Setup action should be available on smtp setup page. did you check?
