Have you checked the IIS authentication methods... both anonymous and windows authentication should be enabled for the CRM site.

we need some more information :

1.How many servers are you using ? Do you have a separate IIS/web server from you SQL server ?

    2. are you trying to connect to MSCRM using the http://Servername.domain.local of the IIS/WEb server or are you using a http://shortname.domain.local / cname / custom redirect ?

    3. Are you using a local account for the IIS app pool (e.g. network service etc) or are you using a service account ?

    4. Have you changed the Internet Explorer security settings to add the address you are using to connect to the MSCRM to 'Trusted Sites' to allow it to present your credentials automatically to iis?

    5. have you done any work so far around the SPN's (service principal names) ?

To work around this problem, enable the Windows Authentication role service in the Web Server (IIS) role.
