In order to conduct in-transit transfers, is it simply a matter of creating an in-transit location and creating a Transfer Order?

That is correct. To simplify this, BC provides Transfer Routes that can be setup to detail the default route from Location A to Location B. For example, in the following setup, whenever BLUE is setup for from location and RED as to location, OUT.LOG will be automatically selected as in-transit location.
Also, you cannot move items to in-transit locations directly, they are marked as in-transit on the location card.
Finally, whenever items are moved as above, transferring them out of BLUE to RED will be a 2 step process moving it from BLUE to OUT.LOG and then from OUT.LOG to RED.
