I've created this subgrid on my opportunity form, however I'd like to make it required so that users must add a record in the subgrid in order to save the opportunity. Is this possible?

Using JavaScript in the form, OnSave check for the number of records in the grid.
(At the time of creation, the number of rows for the subgrid would be 0)

This will be tricky since sub-grids get enabled only when the record is saved. User are allowed to related record by adding them to subgrid of a existing record.
What you can do is probably is show some 'Alerts' on form when user saves the record.

You cant associate any record without successful creation.I think Plugin would be good option and throw Business Process Error as a Notification.
