Is there build in functionality in Dynamics 365 to stop a user from fulfilling an order with no line items?

This functionality is not in Dynamics 365 by default, but you can customise sales order fulfilment so that a user cannot fulfil a sales order with no line items.

I recommend creating a plugin that runs when the sales order is fulfilled. The plugin will need to count the number of sales order lines linked to the parent sales order and throw an InvalidPluginExecutionException if there are no sales order lines. Register the plugin on the create step for the OrderClose entity in the Pre-operation stage.
