I've set up a custom text field on the opportunity form and set the searchable on the field to "Yes", but this field is not searchable. Any ideas?

You have to add the field name to Add Find Columns in Quick Find View for the Entity.

You need to add this field to the "Quick Find" view.
Go to the opportunity entity --> Views ---> Open the "Quick Find" view and add your new field and other fields you require to be searchable.
