I want to migrate from Dynamics 365 On Premise to Dynamics 365 online. Can i do it first on Microsoft dynamics 365 trial version? Or do i need to have license before, even for the testing purpose?

A trial instance has a number of restrictions:-
1) you get a single (production) instance - with a subscription you get (as a minimum) 2 instances (1 production, 1 sandbox for testing / development) and can buy more
2) it lasts 30 days and at some point after that it will be deleted - trials can be extended but it requires asking someone at Microsoft to approve / justify it.
Personally a trial instance is a good way to validate things during the decision phase (such as how much work would be involved in the migration and whether it is possible to achieve what you want online) but you really need to move towards a paid subscription for the migration itself.
If you are concerned about costs you don't need to pay for all the users prior to go-live. You could purchase subscriptions for the development / migration team now and purchase the actual user licences 1-4 weeks before the actual go-live date.
