I have requirement here, when a case is created, i need to call my routing rule set via plugin (customer don't want to click again "Save and Route").
Please help me how can i achieve this

You should use ApplyRoutingRule message - msdn.microsoft.com/.../microsoft.crm.sdk.messages.applyroutingrulerequest.aspx
have you tried ApplyRoutingRuleRequest?
You can do it easly with XrmLibrary
IncidentHelper helper = new IncidentHelper(_organizationService);
helper.ApplyRoute(Guid.Parse("incident record Id"));
or you can use ApplyRoutingRuleRequest like this
ApplyRoutingRuleRequest request = new ApplyRoutingRuleRequest()
Target = new EntityReference("incident", Guid.Parse("incident record Id"))
var response = (ApplyRoutingRuleResponse)_organizationService.Execute(request);
