We're upgrading to GP 2010 and have workflow installed. Will workflow get cleared out and need to be rebuilt with an upgrade? Is there a way to build workflow in an upgraded test environment and retain it when we go live?

Workflow would be upgraded with GP as long as you are not going to be moving to a new SharePoint site. If you want to do this in a test environment it gets a little more complicated. I would recommend you review the Workflow Installation guide which has more detailed upgrade information.
