Is it possible to sort the displayed Document Locations? If so would this affect which default location gets loaded first?

I'm sorry to tell you that there is no way to implement sorting document locations.

While your answer is correct we discovered that if we "ping" the original opportunity after we finished adding the additional document location then the opportunity will be the default first location to load. The document locations seem to be sorted by a last modified date internally. A bit of a hack but it works for now.
