Please let me know which version of IE supports Update Roll Up 12. We had applied roll up 12 on to test server and found that there are some issues. Does this UR require IE 10?

This would be the other way around - you should be on UR10 if you want to use IE10
5.0.9688.583 = RTM
5.0.9688.2730 = UR10
UR12 brings full support for IE10 without using compatibility mode
