Hi there.
Yes, for sure. I'm using it on my local PC and my company has customers who has SQL Server 2008 R2 Express now, on different Windows versions.
Just make sure that all stores and HQ are on the same version of SQL.

Make sure you install the MS SQL Server Backwards compatibility pack as well.
32 bit:
download.microsoft.com/.../SQLServer2005_BC.msi
64bit:
download.microsoft.com/.../SQLServer2005_BC_x64.msi
