How does field mapping work? Does it only work on creation?
Let's say after a record was created, if I entered a value in to a lookup field can other fields automatically be populate if they were set up with field mapping only?

First of all, tell us which D365 product you're using. (D365 consists of many applications, such as Finance and Operations, Sales or Business Central. You're asking in D365 general forum, therefore it's not clear which product you have in mind.)
Then give us some context. There may be many places where fields are mapped in some way for various purposes.
