Are you able to use Alpha Characters in the GL Main Segments. If so, are there any special set up or reporting requirements?

GL accounts are alpahnumeric so you can use alpha characters. If you are going to use wide characters like W then you may want to go to the Account Format Setup window (Tools >> Setup >> Company >> Account Format Setup) and change the display width to one of the other options. If you choose the Help button and go to About this window, you should be able to see the explanation for the different options. This will help so that all the characters will fit within all the GP windows.
