I have created three register in Store Operation Manager and how can set the default register3, so that the POS make the the sales entries on registered3.
Help me about, how to use/handled the different register in Store Operation Manager / POS

Register numbers are assigned in Store Operations Administrator - File - Configuration - Register. Each machine should have a unique register number assigned to it. If you are only selling out of one worksation and want that register to show register number 3, assign it following the path above. The other worksations will need to have numbers 1 and 2 assigned to them.
Once you've assigned a register number, its best to not change it. That way you will know which workstation made what sale based on register number ID.
