Does AX 2012 R3 allow me to send transportation tenders electronically to the desired carriers, or do I have to use the "Print" function and manually send them out? If so, how do I set that up and how does AX "talk" to the carriers?

The tender document is as far as I know not a part of the XML framework supporting TMS.

It would be preferable the tender doc was supported by XML files

When rate, rate shopping or routing functionality is used - it generates XML files for EDI exchanging. You find these under periodic/transactions
