I have some filtering enabled on lookup on case form of Portal. Now when I am trying to render it as drop-down, related filtering is not working on it. Does anyone have a solution for this,

You could refer to my another answer in this thread.
If you want to show the lookup view in drop-down format, you could add a metadata for your lookup field and choose the style to render Lookup as Dropdown.
