Hi all,
as I know
COC Support public and protected method.If required to do modification in PRIVATE method (stnd) using CoC Is it possible?
How I will achieve this?

It's not possible. You need to find another way to implement your change or you can create an extensibility request to MS from LCS support page and ask them to make it protected or add an extensibility point (but usually it takes time).

Then need to raise a request to MS. Then MS will give the final.solution what to do

Yes, but it's not a support request, but an extensibility request where you provide your scenario details and ask to do certain changes in standard code (like make method protected or extract certain code from private method to new protected). MS review the details and provide their decision about the change.
From LCS you can access extensibility request from Support menu
