In standard NAV 2009, how to import data (for example: a column of Item Numbers) from spreadsheet into a purchase order?

You cannot import data from spreadsheet to Purchase Order line from PO. You can use standard tool from: Application Setup->Company Setup->Data Migration
If you need import direct from Excel on purchase order, you can make Dataport with predefined columns e.g.

If you want to insert data in Purchase Order Line, you need to add table 39 (Purchase Line) in Migration Overview. From this line, you need to run Migration->Migration Fields and choouse what fields you want to insert. If you want Item No. e.g., you need to add also: Document Type (from PO header), Buy-from Vendor No. (from PO header), Document No. (from PO header), Line No. (Step-by 10000), Type (Item, Resource, G/L Account, Charge), No. (Item No.)... If you want some other fields, you are free.
After that, you need to run Function->Export to Excel and you will get formated file (for each colimn, you will get comment with field format). In this file, you can fill data manualy, and after that run Function->Import from Excel. System will place these data in Migration Records (temporary table). And now, you need to run Migration->Apply Migration Data.
If data were OK, you are finish. If you have some problem, you can see these data in Migration Errors. You can see error source by press F9. If you change problem in table, system will automatic relocate in destination table (in this case T39).

When exporting to Excel, I received a message: "You do not have the permission to read the Warehouse Class table."
Actually, my user role has all the permissions.
Check whether your license has permissions to read that table..
You try to export data from table not in your license, as Mohana said. Are you realy want data from this table, because you cannot use them. Maybe this is accidentally, because you need to mark table from where you want export data. If you have more tables for migration, please erase them and make only necessary tables.
Highlight or filter only the record of table you are about to export, to avoid the license permission.
