I do not know at which step you are encountering the error but in general Site is mandatory for creating a purchase order from the requisition. Logically as well you need to at least provide site information so that the purchase team can plan purchases accordingly according to the lead times of different suppliers or any other parameters because you need to confirm the delivery schedule from the supplier and site can vastly impact the delivery times from the suppliers and there could be many criteria as well. You just need to add the site on the Purchase requisition lines inventory dimensions tab in the Purchase requisition line for the item.
You can also use dimension display button on the line to display the site directly in the line so that the information is maintained upon adding line items.
Kindly specify the site dimension and then try to complete the Purchase requisition.

You need a site for a purchase order confirmation as I have seen the system allow me to create a requisition without specifying a site in D365 but I believe you are using AX 2012 and I had a similar issue there when we created Purchase requisitions through portal and the system required site to be specified as mandatory. You should not specify it as a default on inventory master as well as there are common items which are used across different sites so keeping a site as a default may lead to some issues if you have multiple users and requisitions across multiple sites. It has to be completed on the run-time and you have approval process afterwards to review the information.
