Hello ,
I am copying PO Lines from Other PO but at that time i want to update a custom Filed value while copying. How can i achieve this ?

You can extend PurchLineCopyFromSource class copy() method. It transfers information from source line to target, there you can update your field.
I am unable to find PurchLineCopyFromSource .
Oh, that's 2012 , this class exists in D365 only. For 2012 modify PurchCopying.copyLine() method.
