Understand we can allocate specific lead times in an item's coverage settings, however if one item can be supplied by multiple vendors (each in varying geographical locations) giving for different lead times, can we make vendor specific lead times for an item?
If so, does this setting override the coverage lead time or attempt to communicate with it?

you can achieve that by setting lead time on the price/discount table.
There is the lead time that has priority over the coverage lead time (so if the vendor have no price specified you will have the coverage lead time).

The information on the price table is more accurate, because you can specify lead time based also on the quantity of your purchase, i.e. if you order more pieces you will have more lead time etc.
This is a suggestion based on my opinion, assuming that you have price list managed for every vendor. There may be a better solution to achieve what you are asking.
