Does anyone know if it is possible to get the GLN field in ship-to-adress mandatory without customization?

    If it is not configured or validated as such, you will have to apply some small customization.

    No its not possible out of the box without customization.

    You need to customise that, not only a mandatory but should also validate the data that is entered in it.
