Hi,
We are using ISV solution in AX 2012 R3. Where we can see the ISV license expiry date and other related information?
Do we really need to pay every year for ISV solution?

HI Hariharan,
Like mentioned above several times, it depends on the ISV vendor. There are some standard ISV licensing options from the AX2012 solution, but ISV vendors can implement their own (additional) security as well. I will also ask you to contact the ISV vendor to get the details you are trying to find.
What you have to pay depends on the contract your company has signed for. It can be a subscription or perpetual license with a yearly enhancement fee.
I believe you are sure that we can’t see ISV license information from AX application, customer source and from partner side.

If you can't find anything that would help, better would be to reach out to ISV. ISV will be able to locate you as a customer and all details behind it. I hope at least you know which ISV.

What next if I couldn’t find any ISV license information in contract?
As Nikolaos mentioned, you can see if there is something from ISV in the system administration module. ISV is the owner and only they can provide you more information. If you can't reach ISV, try reaching out to internal folks who may have received the information when ISV was signed up. The contract documents will have such details.
