Hi,
I am trying to achieve this functionality where in a manager being on leave, the leave/absence request should be forwarded to the super manager.
I tried to use the delegate option in the workflow however I am unable to find any trigger point to the workflow.
The WF that I have configured currently is working absolutely fine for manager approval leaves. Need assistance in configuring the same.

For the bit about 'if a manager hasn't responded within 5 days' bit - check out this article on escalations in workflows which should show you what you need:
It was written for Dynamics AX and it's a few years old now but the workflow editor hasn't changed significantly and it should still apply.
If the manager is on leave they'll need to assign a delegate, and their workflow items can then be auto assigned to the person they nominate. There's more info on this here:

Are you trying to get the workflow to recognise that the manager is on leave and escalate the request to the super manager automatically? Or do you want this to happen if the manager hasn't responded to the request within a certain period?
