Can I open the pdf file which stored in D folder as screenshot below? Is it possible to open the pdf file?

Yes, Default Link option of Dynamics NAV can't open the external file from the Webclient but why don't you use Zetadocs for storing the file
Zetadocs can be access through the Webclient and it can possible to access the attached file even from the Webclient. We have implemented this feature at many of our clients and they are also using the facility through webclient. it is also possible use this through WAN.
